A novel VIPR2 Nomad cell line has been developed stably transfecting cAMP Nomad biosensor and human vasoactive intestinal peptide (VIP) receptor 2 termed VPAC2 into U2OS cell line.

Innoprot cAMP Nomad VIPR2 cell line has been designed to assay compounds or analyze their capability to modulate vasoactive intestinal peptide receptor 1. When the agonist binds to VIPR2 a G protein is activated, which in turn, triggers a cellular response mediated by second messengers (cAMP).

Assay Details

In absence of cAMP, the fluorescent biosensor is localized in the cellular membrane. An increase in this second messenger concentration leads to change in the structural folding of Nomad Biosensor that promotes its cellular relocation in the vesicular trafficking of the cells and a fluorescence increase. Activity is easily quantified on living cells by both image analysis of cytoplasmatic granularity changes and fluorescence intensity.

This highly reproducible assay has been validated using PACAP-38 as a VIPR1 agonist in a High Throughput Screening (HTS) Plarform. This cell line has been validated measuring cAMP increase in the cytosol analyzing Nomad biosensor distribution within the cell and by fluorescence intesity using a microplate reader, showing an average of Z´= 0.74 +/- 0.01 for High Throughput Screening.
