A novel DRD1 Nomad cell line has been developed stably transfecting cAMP Nomad biosensor and Dopamine Receptor D1 termed DRD1 into HEK293 cell line.

Innoprot cAMP-Nomad DRD1 cell line has been designed to assay compounds or analyze their capability to modulate Dopamine Receptor D1. When the agonist binds to DRD1 a G protein is activated, which in turn, triggers a cellular response mediated by second messengers (cAMP).

Assay Details

In absence of cAMP, the fluorescent biosensor is localized in the cellular membrane showing a low fluroescence intensity but when the cAMP concentration is increased, it leads to a change in the structural folding of cAMPNomad Biosensor that promotes its fluorescence intensity. In a cell line co‐expressing the cAMP-Nomad Biosensor and a GPCR, after the receptor activation using its agonist, the activity can be easily quantified on living cells by fluorescence increase signal.

This highly reproducible assay has been validated using Dopamine as a DRD1 agonist in a microplate reader. This cell line has been validated measuring cAMP increase in the cytosol analyzing both fluorescence intensity and Nomad biosensor distribution within the cell, showing an average of Z´= 0.79 +/- 0.01.
