A novel NK3 Nomad cell line has been developed stably transfecting Ca++ Nomad biosensor and human Tachykinin/Neurokinin receptor 3 (NK3) into U2OS cell line.

Innoprot Ca++ Nomad NK3 cell line has been designed to assay compounds or analyze their capability to modulate Tachykinin 3 receptor. When the agonist binds to NK3 a G protein is activated, which in turn, triggers a cellular response mediated by second messengers (Calcium).

Assay Details

In absence of calcium, the fluorescent biosensor is localized in the cellular membrane. An increase in this second messenger concentration leads to change in the structural folding of Nomad Biosensor that promotes its cellular relocation in the vesicular trafficking of the cells. Activity is easily quantified on living cells by image analysis of cytoplasmatic granularity changes.

This highly reproducible assay has been validated using substance P as a NK3 agonist in a High Content Analysis (HCA) Platform. This cell line has been validated measuring calcium increase in the cytosol analyzing Ca++ Nomad biosensor vesicularization, showing an average of Z´= 0.84 +/- 0.02 for High Content Screening.
