A novel TACR2 Nomad Mpx cell line has been developed stably transfecting red fluorescent cAMP Nomad biosensor, green fluorescent Ca++ Nomad biosensor and label free human tachykinin receptor 2 termed TACR2 or NKR2 into U2OS cell line.

Innoprot TACR2 Nomad Mpx cell line has been designed to assay compounds or analyze their capability to modulate tachykinin receptor 2. When the agonist binds to TACR2 a G protein is activated, which in turn,triggers a cellular response mediated by two second messengers (cAMP & Ca++) at the same time.

Assay Details

In absence of Ca++ & cAMP, the fluorescent biosensors are localized in the cellular membrane. An increase in these second messengers concentration leads to change in the structural folding of each respective Nomad Biosensor that promotes its cellular relocation in the vesicular trafficking of the cells. Activity is easily quantified on living cells by both, image analysis of cytoplasmatic granularity changes and fluorescence intensity changes.

This highly reproducible assay has been validated using Neurokinin A as a TACR2 agonist in a High Content Analysis (HCA) Platform for image analysis and in a microplate reader for fluorescence intensity. This cell line has been validated measuring b-arrestin recruitment and Ca++  increase in the cytosol analyzing both Nomad biosensors intensities (red & green), showing averages higher than Z´= 0.81 +/- 0.01 in all determinations.
