A novel CCK2 (CCKBR) Nomad cell line has been developed stably transfecting Arrestin-Nomad biosensor and human Cholecystokinin B receptor (CCK2) into HEK293 cell line.

Innoprot Arrestin-Nomad CCK2 cell line has been designed to assay compounds or analyze their capability to modulate Cholecystokinin B receptor. When an agonist binds to CCKBR a G protein is activated which, in turn, triggers a cellular response mediated by calcium and a subsequent internalization mediated by ß-Arrestin.

Assay Details

In basal conditions, Arrestin Biosensor shows a very low fluorescence intensity. When an agonist binds to CCKBR, the GPCR is phosphorylated by GRKs, promoting the recruitment of arrestin molecules including Arrestin-Nomad Biosensor.

Phosphorylated GPCRs bind to arrestin-Nomad Biosensor, leading to a change in its structural folding and promoting an increase in fluorescence intensity, so ß-Arrestin recruitment is easily quantified on living cells by green fluorescence intensity.

This highly reproducible assay has been validated using CCK Octapeptide, sulfated as a CCK2 agonist in a Multiplate Reader. This cell line has been validated measuring ß-Arrestin recruitment analyzing green fluorescence intensity within the cell, with an EC50 = 1.12x10e-11 M and showing an average of Z´= 0.86 +/- 0.01 for High Throughput Screening.
