Neurite Outgrowth Assay

Neurite outgrowth is a key process during neurogenesis. Complex intracellular signaling is involved in the initiation of neurite protrusion and subsequent elongation. The capacity of neurons to produce and elongate these protrusions can be used as a parameter to measure the capacity of chemical compounds to produce neurogenesis. In another sense the capacity to retract the neurite for a compound could be used as neurotoxic indicator. The neurite outgrowth assay is used to measure neurogenesis or neurotoxicity performing an immunochemical test, dying the specific microtubules (Tuj-1) of neurons.

Neurite Outgrowth Assay Details

In the neurite outgrowth assay the neuronal cultures are incubated with the compound and then performed an immunochemical assay, dying the neuron microtubules with fluorescent antibody antiTuj-1. Images are acquired with the BD Pathway 855 High Content Bioimager and the length and number of neuritis are counted using the AttoVision software and the results are compared with sample controls.
