HiTSeeker are cell lines stably expressing label-free GPCRs, also known as seven-(pass)-transmembrane domain receptors. Each vial of HiTSeeker Muscarinic Acetylcholine Receptor M3 Cell Line contains U2OS cells stably expressing CHRM3 with no tag. HiTSeeker Cell lines proliferate in vitro maintaining GPCR expression following the cell culture manual.

Innoprot HiTSeeker Muscarinic Acetylcholine Receptor M3 Cell Line allows to assay compounds, or analyze their capability to modulate CHRM3. When a ligand binds to the CHRM3, it activates a G protein, which in turn, triggers a cellular response mediated by calcium. HiTSeeker CHRM3 cell line has been validated measuring calcium concentration changes in the cytosol upon activation with oxotremorine. The high reproducibility of this assay allows monitoring CHRM3 receptor activation process in High Throughput Screening.

The muscarinic acetylcholine receptor family consists of five GPCRs that mediate some of the neurotransmission functions of acetylcholine in the CNS and the periphery. The M3 muscarinic receptors appears at many places in the body; such as mooth muscles, both endocrine and exocrine glands, lungs, pancreas and the brain. In the CNS, they induce emesis. Expression of Muscarinic M3 receptors occurs in regions of the brain that regulate insulin homeostasis; such as the hypothalamus and dorsal vagal complex of the brainstem. These receptors are also expressed on pancreatic beta cells and are critical regulators of glucose homoestasis by modulating insulin secretion. In general, they cause smooth muscle contraction and increased glandular secretions.
