The luteinizing hormone\/choriogonadotropin receptor (LHCGR), also lutropin\/choriogonadotropin receptor (LCGR) or luteinizing hormone receptor (LHR) is a transmembrane receptor present predominantly in the ovary and testis, but also in many extragonadal organs such as the uterus and breasts. The receptor interacts with both luteinizing hormone (LH) and chorionic gonadotropins (such as hCG in humans). Its activation is necessary for the hormonal functioning during reproduction. In women, the hormone stimulates the ovaries to produce oestradiol. Two weeks into a woman’s cycle, a surge in luteinizing hormone causes the ovaries to release an egg during ovulation. If fertilization occurs, luteinizing hormone will stimulate the corpus luteum, which produces progesterone to sustain the pregnancy. Both hormones stimulates gonadal function in assisted conception techniques such as in vitro fertilisation (IVF). FluoHiTSeeker are cell lines stably co-expressing label-free GPCRs and the CRE-tGFP reporter gene. Each vial of FluoHiTSeeker LHCGR Cell Line contains HEK293 cells stably expressing human Luteinizing Hormone\/Choriogonadotropin Receptor with no tag; and the CRE-tGFP reporter gene as a biosensor to monitor cAMP concentration changes. FluoHiTSeeker Cell lines proliferate in vitro maintaining GPCR expression following the cell culture manual.<\/p>\n

Innoprot FluoHiTSeeker LHCGR Cell Line allows to assay compounds, or analyze their capability to modulate LHCGR.\u00a0When a ligand binds to the LHCGR, it activates a G protein, which in turn, triggers a cellular response mediated by cAMP. Due to the presence of the CRE-tGFP reporter gene, this cell line allows to monitor cAMP concentration in living cells measuring fluorescence intensity. FluoHiTSeeker LHCGR cell line has been validated measuring cAMP concentration changes in the cytosol upon activation with rhCG. The high reproducibility of this assay allows monitoring FSH receptor activation process in High Throughput Screening.<\/p>\n
