tau is a group of neuronal microtubule-associated proteins that are formed by alternative mRNA splicing. They accumulate in neurofibrillary tangles in Alzheimer’s disease (AD) brain. There is significant evidence that a disruption of normal phosphorylation events results in tau dysfunction. It is a contributing factor to the pathogenic processes of neurodegenerative diseases, such as AD. In this assay, we use a U2OS cell line stably expressing triple mutant (TM) human 0N4R Tau-tGFP. Binding of tau to microtubules and consecuent bundle-formation is phosphorylation dependent. Hyperphosporylation of tau leads to its dissociation from microtubules and aggregation into tangles of paired helical filaments (PHF). On the contrary, kinase inhibitors promote tau binding to microtubules and the formation of microtubule bundles. Quantification of bundles by image analysis allows the identification tau kinase and phosphatase inhibitors.<\/p>\n

Applications<\/strong><\/p>\n tau Phosphorilation Assay Cell Line is an Alzheimer’s Disease in vitro model to identify tau kinase and phosphatase inhibitors and modulators. It consists in U2OS cell line stably expressing human triple mutant Tau-tGFP. To perform the assay,\u00a0cells are incubated with test compounds during 2 hours to evaluate their effects on Tau phosphorylation and binding to microtubules. After incubation, nucleus could be stained with DAPI and cells Tau and MT bundles are detected by fluorescence using image analysis algorithms.<\/p>\n Innoprot offers this assay as a \u201cstable cell line\u201d in cryopreserved vials. Each vial of U2OS cell line stably expressing human triple mutant Tau-tGFP contains 3 million cells and Innoprot provides 2 vials of cells with each order.
