cAMPNOMAD Cells are cell lines stably co-expressing tag-free GPCRs and cAMP Nomad Biosensor to monitor G-protein modulation by Adenylyl cyclase inhibition or stimulation. Each vial of cAMPNOMAD D3 Dopamine Receptor Cell Line contains U2OS cells stably expressing the following constructs:

  • human D3 Dopamine Receptor (DRD3) with no tag
  • cAMP Nomad Biosensor (Green Fluorescence)

cAMPNOMAD DRD3 Cell Line allows to assay compounds analyzing the G-Protein signalling pathway by Go involving receptor activation. When an agonist binds to DRD3, the Go protein is activated which, in turn, triggers a cellular response mediated by cAMP (Adenylyl cyclase inhibition). This leads to an increase of green fluorescence intensity of cAMP biosensor. So, the activity can be easily quantified on living cells in a dose-response manner by fluorescence emission in a microplate reader.

DRD3 encodes the D3 subtype of the dopamine receptor. The D3 subtype inhibits adenylyl cyclase through inhibitory G-proteins. This receptor is present in phylogenetically older regions of the brain, suggesting that this receptor plays a role in cognitive and emotional functions. It is a target for drugs which treat schizophrenia, drug addiction, and Parkinson’s disease. Alternative splicing of this gene results in multiple transcript variants that would encode different isoforms, although some variants may be subject to nonsense-mediated decay (NMD). D3 agonists such as 7-OH-DPAT, pramipexole, and rotigotine, among others, display antidepressant effects in rodent models of depression. DRD3 antagonism has been shown to exacerbate locomotor activity in the brain. cAMPNOMAD D3 Dopamine Receptor Cell Line allows the measurement of the activity of ligands, such as Dopamine or Pramipexole.
