HiTSeeker are cell lines stably expressing label-free GPCRs, also known as seven-(pass)-transmembrane domain receptors. Each vial of HiTSeeker Muscarinic Acetylcholine Receptor M5 Cell Line contains U2OS cells stably expressing CHRM5 with no tag. HiTSeeker Cell lines proliferate in vitro maintaining GPCR expression following the cell culture manual.

Innoprot HiTSeeker Muscarinic Acetylcholine Receptor M5 Cell Line allows to assay compounds, or analyze their capability to modulate CHRM5. When a ligand binds to the CHRM5, it activates a G protein, which in turn, triggers a cellular response mediated by calcium. HiTSeeker CHRM5 cell line has been validated measuring calcium concentration changes in the cytosol upon activation with oxotremorine. The high reproducibility of this assay allows monitoring CHRM5 receptor activation process in High Throughput Screening.

The muscarinic acetylcholine receptor family consists of five GPCRs that mediate some of the neurotransmission functions of acetylcholine in the CNS and the periphery. Muscarinic Acetylcholine Receptor M5 has a discrete expression profile appearing predominantly on dopamine neuron terminals; within the dorsal and ventral striatum that potentiate the release of dopamine and glutamate from midbrain projections. It is the sole mAChR in dopaminergic neurons of the ventral tegmental area and substantia nigra pars compacta. Furthermore, the M₅ mAChR appears densely in the ventral subiculum; which projects to the NAc shell, an input pathway implicated in context-mediated relapse to ethanol seeking.
