Mitochondrial Integrity Assay

Mitochondria are cellular organelles regulating metabolism and cell death pathways. The membrane of the mitochondria is the key component to maintain the cell metabolism and homeostasis. Loss of membrane potential of mitochondria is the previous event of the cell dysfunction; a pathway directly related with the activation of death pathways via caspase 3 activation. Mitochondrial Integrity Assay from Innoprot allows to measure the loss of mitochondrial membrane potential using a fluorescent probe that can predict the cell dysfunction prior to complete cell collapse.

To evaluate the mitochondrial membrane potential (ΔΨm), we use the TMRM fluorescent dye. We incubate the test-compounds with the cells and following we add the TMRM dye, measuring mitochondrial integrity in the automated fluorescent image platform (Cell Insight CX7 HCS Platform). We finally compare the fluorescent signal of test-compounds with the control samples.
