The activity of AVPR1A is mediated by G proteins which stimulate a phosphatidylinositol-calcium second messenger system. AVPR1A receptor belongs to the same subfamily of G-protein coupled receptors of AVPR1B, V2R and OTR receptors. The receptor mediates several actons, such as cell contraction and proliferation, platelet aggregation, release of coagulation factor and glycogenolysis. V1A is coupled to Gq/11 proteins and it appears in a high density on smooth muscle cells. It plays an important role in regulation of blood pressure through arterial vasoconstriction and it stimulates VEGF secretion. AVPR1A also appears in the brain, palying important roles in social behaviours, including affiliation and attachment and related to diseases as autism. HiTSeeker are cell lines stably expressing label-free GPCRs, also known as seven-(pass)-transmembrane domain receptors. Each vial of HiTSeeker AVPR1A Cell Line contains HEK293 cells stably expressing AVPR1A with no tag. HiTSeeker Cell lines proliferate in vitro maintaining GPCR expression following the cell culture manual.
Innoprot HiTSeeker AVPR1A Cell Line allows to assay compounds, or analyze their capability to modulate AVPR1A. When a ligand binds to the AVPR1A, it activates a G protein, which in turn, triggers a cellular response mediated by calcium. HiTSeeker AVPR1A cell line has been validated measuring calcium concentration changes in the cytosol upon activation with AVP. The high reproducibility of this assay allows monitoring AVPR1A receptor activation process in High Throughput Screening.
