_cAMPNOMAD Cells are cell lines stably co-expressing tag-free GPCRs and cAMP Nomad Biosensor to monitor G-protein modulation by cAMP release. Each vial of _cAMPNOMAD MC4R Cell Line contains U2OS cells stably expressing the following constructs:

_cAMPNOMAD MC4R Cell Line allows the assay of compounds analyzing the G-Protein signalling pathway via cAMP production upon receptor activation. When an agonist binds to MC4R, a G protein is activated, which in turn triggers a cellular response mediated by cAMP. This leads to an increase in the green fluorescence intensity of the cAMP biosensor. So, the activity can be easily quantified in living cells in a dose-response manner by fluorescence emission in a microplate reader.

The Melanocortin 4 receptor (MC4R) is a G-protein-coupled receptor (GPCR) encoded by the MC4R gene. It is highly expressed in the paraventricular nucleus of the hypothalamus in humans. The receptor plays a crucial role in the regulation of energy homeostasis, feeding behavior, metabolism, and sexual behavior. MC4R interacts with its primary endogenous ligand, alpha-melanocyte stimulating hormone (α-MSH), a neuropeptide involved in appetite suppression. Upon ligand binding, MC4R activates adenylate cyclase via trimeric Gs proteins, increasing intracellular cAMP levels. This rise in cAMP triggers downstream signaling pathways related to metabolic regulation. _cAMPNOMAD MC4R Cell Line from Innoprot allows the measurement of the activity and potency of ligands, such as α-MSH.

Green Fluorescent cAMP Nomad Biosensor, also expressed in the cell line, enables the monitoring of cAMP concentration changes when receptor is activated/inhibited.
