cAMPNOMAD Cells are cell lines stably co-expressing tag-free GPCRs and cAMP Nomad Biosensor to monitor G-protein modulation via changes in intracellular cAMP levels. Each vial of cAMPNOMAD ADRA2A Receptor Cell Line contains U2OS cells stably expressing the following constructs:
cAMPNOMAD ADRA2A Receptor Cell Line allows to assay compounds analyzing the G-Protein signalling pathway by cAMP involving receptor activation. When an agonist binds to ADRA2A, a G protein is activated, which triggers a cellular response mediated by changes in cAMP levels. The cAMPNOMAD ADRA2A Receptor Cell Line from Innoprot leads to an increase in green fluorescence intensity of the cAMP biosensor. Thus, receptor activity can be easily quantified in living cells in a dose-dependent manner by measuring fluorescence emission with a microplate reader.

The alpha-2A adrenergic receptor (α2A adrenoreceptor); also known as ADRA2A, is a cell membrane-spanning alpha-adrenergic receptor. It interacts with (binds) epinephrine, a hormone and neurotransmitter (ligand synonym, adrenaline). Its signaling usually inhibits adenylate cyclase via Gi protein activation, reducing cAMP levels. However, in the cAMPNOMAD ADRA2A Receptor Cell Line from Innoprot, agonist binding results in an increase in the green fluorescence intensity of the biosensor, reflecting cAMP-related signaling activity. This process mediates physiological responses such as vasoconstriction and modulation of neurotransmitter release. The cAMPNOMAD ADRA2A Receptor Cell Line from Innoprot allows the measurement of the activity and potency of ligands; such as UK-14,304.
Green Fluorescent cAMP Nomad Biosensor also expressed in the cell line, allows monitoring cAMP concentration changes when receptor is activated/inhibited.
