hGR Nuclear Translocation Assay Cell Line

A novel HEK293 Cell line has been developed through stable transfectionof an expression vector for a fusion protein of turboGFP and human Glucocorticoid Receptor. The resultant cell line allows to monitor the dynamics of the hormone/receptor complex in living cells and to assay for compounds inducing tGFP-hGR nuclear translocation and formation of nuclear foci.

Innoprot GR nuclear translocation Cell Line has been designed to assay compounds or analyze stimuli for their ability to modulate glucocorticoid receptor nuclear translocation process and quantifying the fluorescence distribution inside the cells. This highly reproducible assay allows monitoring glucocorticoid receptor nuclear translocation process in High Content Analysis and fluorescence microscope applications.

Each vial of hGR Nuclear Translocation HEK293 Cell Line contains 3 million cells and Innoprot provides two vials of cells with each order. It is guaranteed that these cells will expand in more than 30 passages following the cell culture manual.

hGR Nuclear Translocation Assay Cell Line allows the analyzes of stimuli for their ability to modulate receptor nuclear translocation process. To perform this assay, you have to quantify the fluorescence distribution inside the cells with a High Content Imaging system.

Glucocorticoid receptors (GRs) are nuclear hormone receptors of the NR3C class, which also includes mineralocorticoid, progesterone and androgen receptors. They exist as homodimers coupled to Hsp90 or HMGB proteins, which are shed upon activation. Glucocorticoids exert anti-inflammatory effects principally by repressing expression of the transcription factors AP-1 and NF-κB, and repression of pro-inflammatory genes. GR signaling facilitates an interface between the endocrine stress response and the immune system that is essential for restoring immune homeostasis following a response to stress. Glucocorticoid hormones, such as dexamethasone, cortisol or deoxycorticosterone binds the Glucocorticoid Receptor activating it.
