MPXNOMAD Cells are cell lines stably co-expressing tag-free GPCRs and two Nomad Biosensors to monitor G-protein and β-arrestin modulation. Each vial of MPXNOMAD S1P3 Lysophospholipid Receptor Cell Line contains HEK293 cells stably expressing the following constructs:

  • human S1P3 Lysophospholipid Receptor with no tag
  • cAMP Nomad Biosensor (Red Fluorescence)
  • β-arrestin Nomad Biosensor (Green Fluorescence)

MPXNOMAD S1P3 Lysophospholipid Receptor Cell Line allows to assay compounds analyzing both signalling pathways involving receptor activation; G protein by cAMP flux and β-arrestin recruitment, hence allowing biased activity studies. When an agonist binds to S1P3R a G protein is activated which, in turn, triggers a cellular response mediated by cAMP and a subsequent internalization mediated by ß-Arrestin. This leads to an increase of fluorescence intensity of both β-Arrestin and cAMP biosensors. So, the activity can be easily quantified on living cells in a dose-response manner by fluorescence emission in a microplate reader.

Sphingosine-1-phosphate receptor 3 (S1PR3) gene encodes a member of the EDG family of receptors, part of G protein-coupled receptors. This protein is a functional receptor for sphingosine 1-phosphate and likely contributes to both; the regulation of angiogenesis and also the vascular endothelial cell function. Sphingosine 1-phosphate (S1P) influences heart rate, coronary artery caliber, endothelial integrity, and also lymphocyte recirculation through five related high affinity GPCRs. S1P receptor function in the immune system has experimental advantages in approaching selective S1P receptor function at vascular interfaces. Depletion of S1P3 receptors results in a dramatic inhibition of S1P-induced neurite retraction but, on the contrary results with a significant elongation of neuronal processes in response to S1P.
