TDP-25 Aggregation Assay Cell Line

Innoprot has developed a novel fluorescence cell-based assay cell line for High Content Screening that allows the quantification of pathological TDP-25 aggregates. TDP-25 Aggregation Assay Cell Line consists in a U2OS cell line stably expressing the 25-kDa C-terminal fragment of TDP-43 (a.k.a. TDP-25).

Innoprot offers this cell line as a “stable cell line” in cryopreserved vials. Each vial of U2OS cell line stably expressing fluorescent TDP-25 contains 3 million cells and Innoprot provides 2 vials of cells with each order. These cells proliferate in vitro and they are guaranteed to further expand for more than 30 passages following the instructions provided in the technical sheet.

Amyotrophic lateral sclerosis (ALS) is one of the most common degenerative disease of the motor neuron system. The TDP-43 pathology seems to be a dominant type of pathology across sporadic ALS types. TDP-25 is a C-terminal fragment of TDP-43 containing the aggregation-prone low-complexity domain. Unlike full-length TDP-43, TDP-25 lacks the N-terminal domain, increasing its tendency to misfold and aggregate. This higher aggregation propensity makes the TDP-25 aggregation assay cell line a useful model for studying protein aggregation in experimental assays. To perform the assay just incubate the cells with test compounds and Sodium Arsenite to induce the TDP-25 aggregation. Finally, we quantify TDP-25-tGFP aggregates analizing spot number by image analysis.
