CaNOMAD Cells are cell lines stably co-expressing tag-free GPCRs and Calcium Nomad Biosensor to monitor G-protein modulation by intracellular calcium release. Each vial of CaNOMAD CHRM1 Receptor Cell Line contains U2OS cells stably expressing the following constructs:

  • human Muscarinic Receptor 1 (CHRM1) with no tag
  • Calcium Nomad Biosensor (Green Fluorescence)

CaNOMAD Muscarinic Acetylcholine Receptor M1 Cell Line allows to assay compounds analyzing the G-Protein signalling pathway by Gq/11 involving receptor activation. When an agonist binds to M1 receptor, the Gq/G11 protein is activated which, in turn, triggers a cellular response mediated by calcium via Phospholipase C stimulation. This leads to an increase of green fluorescence intensity of Calcium biosensor. So, the activity can be easily quantified on living cells in a dose-response manner by fluorescence emission in a microplate reader.

The muscarinic acetylcholine receptor family consists of five GPCRs that mediate some of the neurotransmission functions of acetylcholine in the CNS and the periphery. The M1 receptor, along with the M3 and M5 receptors; signal through Gq/11 and subsequent release of Calcium from the ER. The M1 receptor mediates depolarization of ganglia by inhibition of voltage-gated M-type K+ channels. In addition, the M1 receptor mediates venous contraction. The muscarinic cholinergic receptor 1 plays an important role in mediation of vagally-induced bronchoconstriction, but also in the acid secretion of the gastrointestinal tract.
